Baclofen works as a direct agonist at GABA-B receptors. The exact mechanism of action is not fully known. It probably works by inhibiting both monosynaptic and polysynaptic reflexes in the spinal cord by acting as inhibitory neuronal transmitter or by blocking excitatory synaptic transmission through hyperpolarisation of afferent terminals.
Baclofen is used in chronic spasticity as a muscle relaxant.
